Study of ability to destroy DNA and myelin basic protein in immunoglobulin G of blood plasma of multiple sclerosis patients

Purpose. The activity in IgG preparations isolated from blood plasma of multiple sclerosis (MS) patients to hydrolyze DNA and myelin basic protein (MBP) was studied. Discuss the potential of this activity to be a pathogenetic and diagnostic MS marker.
Materials and methods. IgGs were isolated by affine chromatography on protein A-agarose, and their ability to hydrolyze linear form of plasmid DNA and commercial MBP from bovine brain was analyzed.
Results. It was found that some MS patients plasma at the same time had IgG that are able to hydrolyse both MBP and DNA. It was found that one of nine studied IgG preparations was able to hydrolyze both MBP and DNA substrates. This IgG was isolated from blood plasma of 41-year old woman diagnosed for light recidivate form of MS. The IgG of interest was additionally purified by gel filtration on Toyopearl TSK HW-55 F column. The results of that gel filtration in combination with electrophoretic study confirmed that both DNA- and MBP-hydrolyzing activities belong to the IgG, and not to other enzymatic impurities in the preparation.
Conclusions. Thus, in blood of some MS patients there are abzymes (IgG) which can destroy both DNA and MBP.
MS patients IgG preparations differ a lot by their DNA nuclease activity. Perhaps this immunoglobulin enzymatic activity depends on MS patients individual clinical features.
