Structural changes of hippocampus area after experimental ischemia of the brain

Purpose. To study the structural neuronal changes and the glial cell reaction in hippocampal CA1 area at different terms following experimental brain ischemia.
Methods. Brain ischemia was modelled in gerbils by clamping a. carotis on both sides for 7 minutes. The neuronal structure and distribution of immunolabeled glial cells in CA1 area were investigated after 15 min, 2 hours, 1, 3, 7, 14, 30 days following ischemia.
Results. Synaptic terminals structural changes were revealed in early period after reperfusion; the signs of neuronal damage - after 1 day. Delayed neuronal death was observed starting from 3 till 7 day. These events were accompanied by increase of labeled astrocytic and microglial cells number, their activation and spatial redistribution. Glial reaction maximum was observed at day 7-14 after ischemia.
Conclusions. Observed changes in synaptic terminals in CA1 hippocampal area gives an evidence to suggest high level of brain synaptic plasticity induced by ischemia-reperfusion. The delayed neuronal death, reactive astrogliosis and prominent increase in microglial cell number were observed in CA1 hippocampal area at day 3—7 following 7 min of ischemia. Brain ischemia in gerbils provides a suitable model for nervous tissue ischemic damage mechanisms studies and potential neuroprotective agents screening.
